Biological Background: Chk1 Function and Localization
- Serine/threonine-protein kinase (CHEK1 gene) that exists as two isoforms: the full-length kinase (isoform 1) and an endogenous repressor isoform that antagonizes CHK1 to promote S to G2/M transition.
- Required for checkpoint-mediated cell cycle arrest and activation of DNA repair in response to DNA damage or unreplicated DNA. Related references: PMID:11535615 PMID:12399544 PMID:12446774
- Phosphorylates CDC25 family phosphatases (CDC25A, CDC25B, CDC25C) to create 14-3-3 binding sites and inhibit their activity, thus preventing CDK-cyclin activation and blocking cell cycle progression. Related references: PMID:12676583 PMID:12676925 PMID:12759351 PMID:9278511
- Marks CDC25A for proteasomal degradation by phosphorylating multiple serine residues (Ser-76, Ser-124, Ser-178, Ser-279, Ser-293), with Ser-76 priming for NEK11-mediated polyubiquitination. Related references: PMID:12676583 PMID:12676925 PMID:12759351 PMID:19734889 PMID:20090422 PMID:9278511
- Phosphorylates RAD51 at Thr-309, promoting its release from BRCA2 and enhancing chromatin association to facilitate homologous recombination repair. Related references: PMID:15665856
- Phosphorylates TP53 at multiple C-terminal sites, promoting acetylation, transcriptional activation, and cell cycle arrest. Related references: PMID:10673501 PMID:15659650 PMID:16511572
- Additionally phosphorylates FANCE (DNA cross-link repair), TLK1 (chromatin assembly), and other substrates to coordinate genome maintenance. Related references: PMID:17296736 PMID:12660173 PMID:12955071
- Localizes to the nucleus, chromosomes, cytoplasm, and centrosome; ubiquitously expressed with highest levels in thymus, testis, small intestine, and colon.
Experimental Guidance and Technical Tips
- To confirm phospho-specificity, treat samples with lambda phosphatase or include a non-phosphorylated peptide block control.
- For Western blot, stimulate cells with DNA-damaging agents (e.g., etoposide, UV, or hydroxyurea) to induce Chk1 phosphorylation at S317 and enhance signal detection.
- Validate antibody performance in the relevant human cell lines and assay systems, as cross-reactivity to other species has not been documented.
- For ELISA applications, employ appropriate positive and negative controls to ensure specificity for the phosphorylated epitope.
- Consider optimizing antibody dilution and incubation conditions for your specific experimental setup.
