Biological Background: Serine/threonine-protein kinase Chk1 Function and Localization
- Serine/threonine-protein kinase Chk1 (CHEK1) is a critical kinase for checkpoint-mediated cell cycle arrest and DNA repair. An alternatively spliced isoform (isoform 2) acts as an endogenous repressor of isoform 1, antagonizing its function.
- Required for the DNA damage response: recognizes the substrate consensus sequence [R-X-X-S/T] and phosphorylates CDC25A, CDC25B, and CDC25C, creating 14-3-3 binding sites that inhibit these phosphatases and/or promote CDC25A degradation, resulting in inhibitory tyrosine phosphorylation of CDK-cyclin complexes and cell cycle arrest. - Phosphorylates RAD51 at Thr-309, promoting its release from BRCA2 and enhancing chromatin association, thereby facilitating DNA repair by homologous recombination. Related references: PMID:15665856

Experimental Guidance and Technical Tips
- The immunogen is a phospho-peptide surrounding Ser280 of human Chk1. The antibody is designed to recognize the phosphorylated form specifically. Validate specificity by comparing signals in phosphatase-treated vs. untreated samples or using a non-phosphorylated blocking peptide.
- For Western blot, using appropriate phosphatase inhibitors during sample preparation is recommended to preserve the phosphorylation signal. A band near 54 kDa is expected for Chk1.
- In ELISA, consider using a plate-bound phospho-peptide and a non-phospho peptide control to confirm phospho-specificity.
- Chk1 phosphorylation at Ser280 is induced by DNA damage. Stimulating cells with DNA-damaging agents (e.g., UV, hydroxyurea) may enhance detection in relevant cell lines.
