Biological Background: c-Kit Function and Localization
- KIT (CD117/c-Kit) is a receptor tyrosine kinase for stem cell factor (KITLG/SCF), playing critical roles in hematopoiesis, gametogenesis, and melanogenesis.
- Functions in cell survival, proliferation, stem cell maintenance, and mast cell development, migration, and function.
- Activates multiple signaling cascades including PI3K/AKT, RAS/RAF/MAPK, and JAK/STAT pathways.
- Phosphorylation at Tyr721 (corresponding to Tyr719 in mouse) is a key autophosphorylation event that facilitates binding and activation of downstream signaling molecules.
- Expressed in testis in spermatogonia, interstitial Leydig cells, and ejaculated spermatozoa. Related references: PMID:20601678
- Subcellularly localized predominantly at the cell membrane, with some cytoplasmic presence.

Experimental Guidance and Technical Tips
- The antibody was raised against a synthetic phosphopeptide corresponding to residues surrounding Tyr719 of mouse Kit. Cross-reactivity with human and rat is expected due to high sequence homology.
- For Western blot, a band around 110–145 kDa (mature glycosylated form) may be detected; consider using appropriate stimulation conditions to induce tyrosine phosphorylation.
- ELISA may be used for detecting phospho-c-Kit in cell lysates; validate with positive and negative controls.
