Biological Background: Mast/stem cell growth factor receptor Kit Function and Localization
- KIT, also known as proto-oncogene c-Kit, CD117, or mast/stem cell growth factor receptor, is a receptor tyrosine kinase encoded by the KIT gene.
- It functions as the cell-surface receptor for the cytokine stem cell factor (SCF/KITLG) and is essential for regulating cell survival, proliferation, hematopoiesis, stem cell maintenance, gametogenesis, mast cell development, migration, and melanogenesis.
- Upon ligand binding, KIT activates multiple downstream signaling pathways, including PI3K/AKT, RAS/RAF/MAPK, and JAK/STAT cascades, resulting in the phosphorylation of various substrates such as PIK3R1, PLCG1, CBL, and members of the STAT family.
- The receptor is primarily localized to the cell membrane and can also be found in the cytoplasm.
- At the protein level, KIT is detected in spermatogonia and interstitial Leydig cells in the testis, and its expression is maintained in ejaculated spermatozoa.
