Biological Background: ABL1 Function and Localization
- ABL1 is a non-receptor tyrosine kinase involved in diverse processes including actin remodeling, cell adhesion, receptor endocytosis, autophagy, DNA damage response, and apoptosis.
- Phosphorylation of WASF3 by ABL1 is critical for lamellipodia formation and cell migration. Related references: PMID:22810897
- In the nucleus, ABL1 participates in DNA repair and apoptotic responses, phosphorylating substrates such as TP73 and CASP9.
- The protein shuttles between the cytoplasm, cytoskeleton, nucleus, and mitochondria, with nuclear translocation linked to DNA damage and mitochondrial targeting linked to oxidative stress.

Experimental Guidance and Technical Tips
- This phospho-specific antibody was raised against a synthetic phosphopeptide corresponding to residues surrounding Tyr245 of human ABL1. It is recommended to treat cell or tissue lysates with phosphatase inhibitors to preserve phosphorylation.
- For Western blotting, the predicted molecular weight is approximately 123 kDa; validate the band in stimulated vs. unstimulated samples or treat with phosphatases as a specificity control.
- In IHC-P, antigen retrieval and phosphatase inhibitor pretreatment of tissue sections may be required to retain the phospho-epitope.
- Consider using ELISA with phosphopeptide and non-phosphopeptide controls to confirm phospho-specificity.
