Biological Background: Bcl-2 Function and Localization
- Apoptosis regulator Bcl-2, encoded by the BCL2 gene, is also known as PPP1R50.
- Suppresses apoptosis in a variety of cell systems, including factor-dependent lymphohematopoietic and neural cells. Related references: PMID:1508712 PMID:8183370
- Regulates cell death by controlling mitochondrial membrane permeability and functions in a feedback loop with caspases; inhibits caspase activity by preventing cytochrome c release or binding APAF-1. Related references: PMID:11368354
- Acts as an inhibitor of autophagy by interacting with BECN1 and AMBRA1 under non-starvation conditions. Related references: PMID:18570871 PMID:20889974 PMID:21358617
- May attenuate inflammation by impairing NLRP1-inflammasome activation, thus reducing CASP1 activation and IL1B release. Related references: PMID:17418785
- Subcellular localization includes mitochondrion outer membrane, nucleus membrane, endoplasmic reticulum membrane, and cytoplasm.
- Tissue specificity: expressed in a variety of tissues.
- Subject to phosphorylation (e.g., Thr56), alternative splicing, and Ubl conjugation; frequently rearranged in B-cell lymphomas.
Experimental Guidance and Technical Tips
- When performing western blot, include phosphatase inhibitors in the lysis buffer to preserve the T56 phosphorylation state.
- Validate antibody specificity using positive controls such as cell lines or tissues known to express Bcl-2 under conditions that induce Thr56 phosphorylation.
- For IHC-P, optimize antigen retrieval and consider phospho-specific detection conditions to ensure specific staining.
- In ELISA, a phosphorylated peptide corresponding to the immunogen can serve as a positive control to confirm assay sensitivity.
