Biological Background: ATF-2 Function and Localization
- ATF-2 (Activating transcription factor 2), also known as CREB-2, CRE-BP1, and HB16, is a member of the ATF/CREB family of basic region-leucine zipper (bZIP) transcription factors, encoded by the ATF2 gene.
- Functions as a transcriptional activator by binding to CRE (5'-TGACGTCA-3') or AP-1 (5'-TGACTCA-3') consensus sequences, regulating genes involved in anti-apoptosis, cell growth, and DNA damage response.
- Exhibits intrinsic histone acetyltransferase (HAT) activity, specifically acetylating histones H2B and H4 in vitro, contributing to chromatin modification and transcriptional regulation. Related references: PMID:10821277
- This phosphorylation-specific antibody detects ATF2 phosphorylated at Thr69 and Thr71 by kinases such as ATM, which is critical for its role in the DNA damage response, ionizing radiation-induced S phase checkpoint, and recruitment of the MRN complex to DNA damage foci.
- In the cytoplasm, ATF2 localizes to the mitochondrial outer membrane where it interacts with HK1 and VDAC1, impairing mitochondrial membrane potential and promoting cell death, highlighting its dual role in transcription and apoptosis.
- The protein is ubiquitously expressed, with higher levels in the brain, and undergoes nuclear–cytoplasmic shuttling; its subcellular localization includes the nucleus, cytoplasm, and mitochondrion outer membrane.
- ATF2 can have either oncogenic or tumor suppressor activities depending on tissue context, and it modulates ATM activity by promoting KAT5 degradation in concert with CUL3 and RBX1.
Experimental Guidance and Technical Tips
- This antibody was raised against a synthetic phosphopeptide corresponding to Thr69 and Thr71 of human ATF2. For phosphorylation-specific detection, validate specificity by treating cell lysates with lambda phosphatase or using non-phosphorylated peptide competition.
- For Western blotting, the expected molecular weight is approximately 55 kDa. Use appropriate blocking and washing conditions, and consider including a phosphatase inhibitor cocktail in lysis buffers to preserve phosphorylation.
- For immunohistochemistry (IHC-P), antigen retrieval may be required; optimization of antibody dilution and detection system is recommended for each tissue type. Include appropriate positive and negative controls.
- Cross-reactivity with mouse and rat ATF2 allows the antibody to be used in rodent models, but sequence alignment and pilot experiments are advised to confirm reactivity.
