Biological Background: PHLPP1 Function and Localization
- PHLPP1 (PH domain leucine-rich repeat-containing protein phosphatase 1) is a Ser/Thr protein phosphatase that specifically dephosphorylates the hydrophobic motif of AGC kinases, including 'Ser-473' of AKT2 and AKT3, leading to apoptosis and tumor growth suppression. Related references: PMID:15808505 | PMID:17386267 | PMID:18162466
- It also dephosphorylates PRKCA and PRKCB, causing their destabilization and degradation, and acts on other targets like STK4 and RPS6KB1 to regulate apoptosis and translation. Related references: PMID:18162466 | PMID:20513427 | PMID:21986499
- PHLPP1 functions as a tumor suppressor, inhibiting cancer cell proliferation, and dephosphorylates RAF1 to inhibit its kinase activity. Related references: PMID:19079341 | PMID:24530606
- The protein contains a PH domain and leucine-rich repeats, and is expressed in multiple subcellular compartments, including cytoplasm, nucleus, and cell membrane.
- It is ubiquitously expressed in non-cancerous tissues, but expression is often lost or decreased in colorectal tumors, and isoform 2 plays a major role in hippocampal neurons.
- PHLPP1 is involved in circadian rhythm regulation and hippocampus-dependent long-term memory formation.
- Post-translational modifications include acetylation and phosphorylation; it is alternatively spliced to generate multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen was designed against a C-terminal recombinant fragment (aa 1160-1428). Verify reactivity in your specific experimental system, as endogenous protein context may influence epitope accessibility.
- For Western blot (WB), the predicted band is approximately 185 kDa; use appropriate molecular weight markers and consider a high-percentage gel or gradient gel for optimal resolution.
- For IHC-P, antigen retrieval methods should be optimized; include proper positive and negative control tissues to confirm specificity.
- For IF/ICC, fixation and permeabilization conditions may affect staining; test different protocols to achieve optimal signal-to-noise ratio.
- The antibody is reported to cross-react with human, mouse, and rat PHLPP1; validate in relevant species samples if used beyond these.
