- The protein pleckstrin homology-like domain family A member 2 (PHLDA2), also known as IPL, TSSC3, or BWR1C, is a 17 kDa cytoplasmic and membrane-associated protein containing a pleckstrin homology (PH) domain.
- PHLDA2 plays a critical role in regulating placenta growth by competing with other PH domain-containing proteins for membrane lipid binding, thereby modulating signaling pathways (By similarity).
- It is highly expressed in the placenta, particularly in all cells of the villous cytotrophoblast, and at low levels in adult liver, lung, and fetal liver; expression is also detected in adult prostate gland and brain, as well as in neuroblastoma, medulloblastoma, and glioblastoma cell lines.
- PHLDA2 is an imprinted gene, with expression largely from the maternal allele, and its protein is absent in hydatidiform moles, linking it to abnormal trophoblast development and imprinted gene disorders.
- The protein is classified as a phosphoprotein and has been identified through proteomics studies; it carries a reference proteome designation from UniProt.
- Alternative names such as tumor-suppressing STF cDNA 3 protein highlight its potential involvement in growth suppression, consistent with its downregulation in certain cancers.
