Biological Background: PHD3 Function and Localization

  • PHD3, also known as Egl nine homolog 3, HIF-prolyl hydroxylase 3 (HIF-PH3), and prolyl hydroxylase domain-containing protein 3 (PHD3), is encoded by the EGLN3 gene and is a prolyl hydroxylase that mediates hydroxylation of proline residues in target proteins such as PKM, TELO2, ATF4, GPX4, and HIF1A (PubMed:19584355, PubMed:20978507, PubMed:21483450).   • As a cellular oxygen sensor, under normoxic conditions PHD3 catalyzes hydroxylation of specific proline residues in hypoxia-inducible factor alpha (HIF1A and HIF2A), targeting them for proteasomal degradation via the von Hippel-Lindau complex, thereby attenuating the hypoxic response (PubMed:11595184, PubMed:12181324).
