Biological Background: PGM2 Function and Localization
- Phosphopentomutase (PGM2), also known as glucose phosphomutase 2, phosphodeoxyribomutase, and phosphoglucomutase-2, is encoded by the PGM2 gene in humans.
- Functions as a phosphopentomutase that catalyzes the interconversion of ribose-1-phosphate and deoxyribose-1-phosphate to their corresponding 5-phosphopentoses, playing a key role in nucleotide salvage pathways. Related references: PMID:17804405
- Also exhibits phosphoglucomutase activity, reversibly isomerizing α-D-glucose 1-phosphate to α-D-glucose 6-phosphate, albeit with lower catalytic efficiency, via an α-D-glucose 1,6-bisphosphate intermediate. Related references: PMID:17804405
- Possesses weak glucose 1,6-bisphosphate synthase activity in vitro, though this is unlikely to be physiologically relevant. Related references: PMID:17804405, PMID:18927083
- Primarily localized to the cytoplasm and cytosol.
- Subject to post-translational modifications including acetylation and phosphorylation; requires magnesium as a cofactor.
- Alternatively spliced variants have been reported.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1-130) of human PGM2, which may be suitable for detecting full-length and N-terminal isoforms.
- For Western blotting, consider using whole-cell lysates from human or mouse tissues/cell lines expressing PGM2; the theoretical molecular weight is approximately 68 kDa.
- For ELISA applications, validate with recombinant PGM2 protein or peptide to ensure specificity and sensitivity.
