  • PGLYRP2, also known as peptidoglycan recognition protein 2 or PGRP-L, is a member of the peptidoglycan recognition protein family, which plays a crucial role in innate immunity.
  • It functions as an N-acetylmuramoyl-L-alanine amidase, hydrolyzing the amide bond between N-acetylmuramic acid and L-alanine residues in bacterial peptidoglycan.
  • The protein acts as a scavenger by digesting biologically active peptidoglycan (PGN) into inactive fragments, thereby modulating immune responses, but has no direct bacteriolytic activity.
  • Alternative splicing generates multiple isoforms; isoform 2 is not detected in the liver or serum, suggesting tissue-specific regulation.
  • PGLYRP2 is strongly expressed in the liver and fetal liver, and is secreted into the serum; lower expression is observed in transverse colon, lymph nodes, heart, thymus, pancreas, descending colon, stomach, and testis.
  • Subcellularly, the protein is secreted and also found associated with membranes, consistent with its role in extracellular peptidoglycan processing.
  • The protein contains key structural features including a signal peptide, a zinc-binding domain essential for its amidase activity, disulfide bonds, and glycosylation sites; it is also regulated by phosphorylation.

  • The immunogen is a synthetic peptide from amino acids 300–400 of human PGLYRP2; therefore, the antibody may detect multiple isoforms containing this region.
  • For Western blot (WB), the predicted molecular weight is ~62 kDa, but due to glycosylation and phosphorylation, the observed band may appear at a higher or different molecular weight; validate with appropriate controls.
  • This antibody cross-reacts with mouse PGLYRP2, making it suitable for studies in mouse models; however, validate specificity in the relevant tissue or cell lysate.
  • For ELISA applications, consider using the immunizing peptide as a positive control and titrate the antibody to determine optimal working dilutions.
