Biological Background: PGC1α/β Function and Localization

  • PGC1α (PPARGC1A) and PGC1β (PPARGC1B) are homologous transcriptional coactivators. PGC1α is also known as ligand effect modulator 6 (LEM6) and exists in multiple isoforms due to alternative promoter usage and splicing. The predicted molecular weight for PGC1α is ~91 kDa, but post-translational modifications may result in higher observed weights (e.g., 113 kDa).
  • Acts as a transcriptional coactivator for steroid receptors and nuclear receptors, significantly enhancing the transcriptional activity of PPARG and thyroid hormone receptor on the uncoupling protein (UCP1) promoter. Related references: PMID:10713165 PMID:20005308 PMID:21376232
  • Regulates key mitochondrial genes and drives adaptive thermogenesis, playing a central role in mitochondrial biogenesis and energy expenditure. Related references: PMID:10713165 PMID:20005308 PMID:21376232
  • Essential for metabolic reprogramming in response to dietary availability, coordinating the expression of a wide array of genes involved in glucose and fatty acid metabolism. Related references: PMID:10713165 PMID:20005308 PMID:21376232
  • Key regulator of gluconeogenesis: stimulates hepatic gluconeogenesis by increasing the expression of gluconeogenic enzymes and acts together with FOXO1 to promote the fasting gluconeogenic program. Related references: PMID:16753578 PMID:23142079
  • Induces the expression of PERM1 in skeletal muscle in an ESRRA-dependent manner, linking PGC1α to muscle energy metabolism. Related references: PMID:23836911
  • Integrates circadian rhythms with energy metabolism, required for oscillatory expression of clock genes (BMAL1, NR1D1) through coactivation of RORA and RORC, and metabolic genes (PDK4, PEPCK). (By similarity)
  • Subcellular localization: Predominantly nuclear, found in PML bodies, and also detected in the cytoplasm. Tissue specificity: Highly expressed in heart, skeletal muscle, liver, and kidney; lower levels in brain, pancreas, and white adipose tissue. Expression is modulated by obesity and fasting.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide derived from a region shared by human PGC1α and PGC1β (aa 699-798), suggesting the antibody may recognize both paralogs. Verify specificity in the relevant sample system.
  • For Western blotting, use a low-percentage polyacrylamide gel (e.g., 6-8%) and optimize transfer conditions for high-molecular-weight targets (113 kDa). Consider using a positive control lysate from PGC1α/β-rich tissues (e.g., liver, skeletal muscle).
  • For ELISA applications, titrate the antibody in direct or capture formats and validate using recombinant PGC1α/β protein or cell lysates from stimulated cells.
  • The reported cross-reactivity with mouse and rat makes this antibody suitable for preclinical models; however, confirm cross-reactivity experimentally by comparing human, mouse, and rat samples where possible.
  • To study nuclear/cytoplasmic shuttling, fractionate cells and assess PGC1α/β distribution under baseline and stimulated conditions (e.g., fasting, exercise).
