Biological Background: PGC1α/β Function and Localization
- PGC1α (PPARGC1A) and PGC1β (PPARGC1B) are homologous transcriptional coactivators. PGC1α is also known as ligand effect modulator 6 (LEM6) and exists in multiple isoforms due to alternative promoter usage and splicing. The predicted molecular weight for PGC1α is ~91 kDa, but post-translational modifications may result in higher observed weights (e.g., 113 kDa).

- For Western blotting, use a low-percentage polyacrylamide gel (e.g., 6-8%) and optimize transfer conditions for high-molecular-weight targets (113 kDa). Consider using a positive control lysate from PGC1α/β-rich tissues (e.g., liver, skeletal muscle).
- For ELISA applications, titrate the antibody in direct or capture formats and validate using recombinant PGC1α/β protein or cell lysates from stimulated cells.
- The reported cross-reactivity with mouse and rat makes this antibody suitable for preclinical models; however, confirm cross-reactivity experimentally by comparing human, mouse, and rat samples where possible.
