Biological Background: PGC1α Function and Localization
- PGC1α, encoded by PPARGC1A, is a transcriptional coactivator that potently enhances the activity of PPARG and thyroid hormone receptor on the uncoupling protein promoter, regulating adaptive thermogenesis. Related references: PMID:10713165, PMID:20005308, PMID:21376232
- It orchestrates metabolic reprogramming in response to nutrient availability by coordinating the expression of genes involved in glucose and fatty acid metabolism. Related references: PMID:10713165, PMID:20005308, PMID:21376232
- A critical regulator of hepatic gluconeogenesis: PGC1α increases the expression of gluconeogenic enzymes and cooperates with FOXO1 to promote the fasting gluconeogenic program. Related references: PMID:16753578, PMID:23142079
- In skeletal muscle, PGC1α induces the expression of PERM1 in an ESRRA-dependent manner, contributing to muscle oxidative capacity. Related references: PMID:23836911
- PGC1α integrates circadian rhythms with energy metabolism by coactivating RORA and RORC, driving oscillatory expression of clock genes (BMAL1, NR1D1) and metabolic genes (PDK4, PEPCK).
- It localizes predominantly to the nucleus and PML bodies, but can also be found in the cytoplasm under certain conditions.
- Tissue expression is highest in heart, skeletal muscle, liver, and kidney; lower in brain and pancreas; and very low in intestine and white adipose tissue. In skeletal muscle, PGC1α levels are reduced in obesity and induced by fasting.
- Post-translational modifications include acetylation, phosphorylation, and ubiquitylation, and multiple isoforms are produced by alternative promoter usage and alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 340-480 of human PGC1α, a region likely to be exposed in the native protein. Consider using this antibody for Western blot detection of PGC1α ~91 kDa in human cell or tissue lysates. Validate specificity with appropriate positive and negative controls.
- For ELISA applications, this antibody may be used as a capture or detection reagent; pairing with a validated antibody pair is recommended for optimal sensitivity.
- The predicted molecular weight is 91 kDa; ensure adequate gel percentage and transfer conditions for high-molecular-weight proteins.
