Biological Background: PGC1 alpha Function and Localization
- PGC1 alpha (peroxisome proliferator-activated receptor gamma coactivator 1-alpha) is a transcriptional coactivator that interacts with nuclear receptors, notably PPARG and thyroid hormone receptor, to enhance the transcription of genes involved in energy metabolism. Related references: PMID:12754525 PMID:15744310 PMID:23217713
- Functions as a master regulator of mitochondrial biogenesis and oxidative metabolism, and plays an essential role in adaptive thermogenesis. Related references: PMID:12754525 PMID:15744310 PMID:23217713
- Orchestrates metabolic reprogramming in response to dietary availability by coordinating genes involved in glucose and fatty acid metabolism. Related references: PMID:12754525 PMID:15744310 PMID:23217713
- Acts as a key regulator of gluconeogenesis in the liver, stimulating the expression of gluconeogenic enzymes in cooperation with FOXO1 to promote the fasting response. Related references: PMID:12754525
- Participates in the integration of circadian rhythms with energy metabolism through coactivation of RORA and RORC, and is required for oscillatory expression of clock and metabolic genes. Related references: PMID:17476214
- Predominantly localizes to the nucleus, particularly within PML bodies, where it exerts its coactivator functions.
- Highly expressed in tissues with high mitochondrial content, including skeletal muscle, brown adipose tissue, heart, kidney, and brain.
- Subject to post-translational modifications such as acetylation, phosphorylation, and ubiquitination, which modulate its activity and stability.
Experimental Guidance and Technical Tips
- The immunogen comprises amino acids 1–300 of mouse PGC1 alpha, a region near the N-terminus; this design may recognize multiple isoforms or cleavage products—validate specificity in your experimental system.
- For Western blotting, expect a main band around 91 kDa in lysates from PGC1 alpha-rich tissues (skeletal muscle, brown adipose tissue, heart); additional bands may occur.
- In immunofluorescence and immunohistochemistry, nuclear staining is anticipated; optimize fixation and permeabilization conditions, and include appropriate controls.
- The antibody is suitable for ELISA; titrate to determine the optimal working concentration for your assay.
