Biological Background: PGAM5 Function and Localization
- PGAM5 is a serine/threonine-protein phosphatase primarily located in mitochondria, existing as multiple isoforms from alternative splicing. It is also known as BXLBV68 and belongs to the phosphoglycerate mutase family.
- It resides on both the outer and inner mitochondrial membranes, a unique dual topology critical for its diverse functions.
- PGAM5 regulates mitochondrial fission by dephosphorylating DNM1L/DRP1, thereby modulating cellular senescence. Related references: PMID:32439975
- It dephosphorylates MFN2, preventing its ubiquitination and degradation to maintain mitochondrial network integrity. Related references: PMID:37498743
- PGAM5 promotes PARKIN-independent mitophagy by interacting with and dephosphorylating FUNDC1, facilitating its association with LC3. Related references: PMID:24746696
- It participates in the anti-oxidative response by forming a complex with KEAP1 and NRF2. Related references: PMID:18387606
- PGAM5 is a key mediator of necroptosis, acting as a substrate for RIPK3 and recruiting the RIPK1-RIPK3-MLKL necrosome to mitochondria. Related references: PMID:22265414
Experimental Guidance and Technical Tips
- The immunogen corresponds to a region within the human PGAM5 protein; verify sequence similarity with target species to ensure expected cross-reactivity.
- For Western blotting, the predicted protein weight is 32 kDa, but actual band position may vary due to isoforms or modifications; validate with appropriate controls.
- For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), mitochondrial localization may require optimized permeabilization and fixation protocols.
- In ELISA applications, titrate antibody and antigen concentrations to achieve optimal signal-to-noise ratio.
