Biological Background: Pepsin A-5 Function and Localization
- Pepsin A-5 (pepsinogen-5) is an aspartyl protease encoded by the PGA5 gene in humans. It belongs to the peptidase A1 family of pepsin-like enzymes.
- The enzyme exhibits broad substrate specificity, preferentially cleaving peptide bonds involving phenylalanine and leucine, but also hydrolyzing many other bonds.
- It is secreted as an inactive zymogen (pepsinogen) and activated by autocatalytic cleavage under acidic conditions, releasing the active pepsin A-5.
- Pepsin A-5 functions primarily in protein digestion in the stomach, where it contributes to the breakdown of dietary proteins into peptides.
- Subcellular localization is extracellular, being secreted into the gastric lumen.
- Post-translational modifications include phosphorylation and formation of disulfide bonds, which are critical for structural stability.
- Direct protein sequencing evidence supports its existence, and it has been identified in proteomics studies.
Experimental Guidance and Technical Tips
- The antibody is generated against a recombinant fusion protein covering amino acids 63–180 of human PGA5. This region lies within the pepsin moiety, so the antibody may recognize both the zymogen (pepsinogen-5) and active pepsin A-5 forms. Users should validate specificity in their own sample system.
- For Western blot, expected bands may appear around 42 kDa for the full-length protein; however, due to zymogen processing, additional bands of lower molecular weight could be observed. Appropriate positive controls and molecular weight markers are recommended.
- ELISA applications may benefit from optimizing coating antigen and blocking conditions. Cross-reactivity with other pepsinogen family members should be assessed if required.
