- PFKM encodes the muscle-specific isoform of ATP-dependent 6-phosphofructokinase, also known as phosphofructo-1-kinase isozyme A (PFK-A) or phosphohexokinase.
- It catalyzes the phosphorylation of D-fructose 6-phosphate to fructose 1,6-bisphosphate, the first committing step of glycolysis, using ATP as a phosphate donor.
- This enzyme is an allosteric regulator of glycolysis, responding to cellular energy status, and is critical for muscle ATP production.
- Mutations in PFKM cause glycogen storage disease type 7 (Tarui disease), an autosomal recessive disorder characterized by exercise-induced muscle cramps and myoglobinuria (disease variant).
- PFKM localizes to the cytoplasm and undergoes multiple post-translational modifications, including phosphorylation, acetylation, hydroxylation, and glycosylation.
