Biological Background: PEX5 Function and Localization
- PEX5, also known as peroxisomal targeting signal 1 receptor (PTS1 receptor), PTS1-BP, or peroxin-5, is a key component of the peroxisomal protein import machinery, existing in multiple isoforms via alternative splicing.
- Mediates peroxisomal import of proteins containing a C-terminal PTS1-type tripeptide peroxisomal targeting signal (SKL-type) by binding to cargo proteins in the cytosol and translocating them through the PEX13-PEX14 docking complex into the peroxisome matrix. - Following cargo translocation, PEX5 is retrotranslocated into the cytosol in a process that leads to release of bound cargo in the peroxisome matrix, resetting the receptor for subsequent import cycles. Related references: PMID:11336669 PMID:24662292
- In addition to PTS1 cargo, PEX5 mediates peroxisomal import of proteins containing a C-terminal PTS2-type targeting signal by binding to the PEX7 receptor when it is associated with PTS2-cargo proteins, thereby bridging the PTS2-cargo-PEX7 complex to the peroxisomal docking machinery.
