Biological Background: PEX2 Function and Localization
- PEX2, also known as peroxin-2, PMP35, or PAF-1, is a 35 kDa peroxisomal membrane protein and member of the peroxin family.
- PEX2 is an essential E3 ubiquitin-protein ligase that forms the retrotranslocation channel with PEX10 and PEX12 to export PEX5 from peroxisomes, enabling PEX5 recycling (PubMed:24662292). Related references: PMID:24662292
- PEX2 mediates pexophagy by ubiquitinating peroxisomal proteins such as PEX5 and ABCD3/PMP70 in response to starvation (PubMed:27597759). Related references: PMID:27597759
- In response to reactive oxygen species, PEX2 catalyzes Lys-48-linked polyubiquitination and degradation of PNPLA2/ATGL, regulating lipolysis (PubMed:34903883). Related references: PMID:34903883
- It localizes exclusively to the peroxisome membrane as a multi-pass transmembrane protein.
- Mutations in PEX2 are associated with peroxisome biogenesis disorders, including Zellweger syndrome spectrum (PBD5A, PBD5B).
- PEX2 contains a RING-type zinc finger domain, characteristic of its E3 ubiquitin ligase activity, and is regulated by acetylation and disulfide bond formation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1–130, the N-terminal region of human PEX2. This polyclonal antibody may recognize full-length PEX2 and potential isoforms; validation in the relevant sample system is recommended.
- For Western blot (WB), the predicted molecular weight of PEX2 is approximately 35 kDa (34.8 kDa by calculation). A dilution range of 1:500–1:2000 is commonly used for rabbit polyclonal antibodies; optimize for your specific sample lysates.
- In immunohistochemistry (IHC-P), antigen retrieval using citrate or EDTA buffer may enhance detection; confirm with appropriate controls.
- For ELISA, the antibody can be used for detection of recombinant or endogenous PEX2; consider paired antibody combinations for sandwich ELISA.
