Biological Background: PEX12 Function and Localization
-
PEX12, also known as Peroxin-12 or PAF-3, is a component of the peroxisomal retrotranslocation channel that, together with PEX2 and PEX10, mediates the export of the PEX5 receptor from the peroxisomal membrane to the cytosol, enabling its recycling (PubMed:24662292, PubMed:9354782, PubMed:9632816). Related references: PMID:24662292 PMID:9354782 PMID:9632816
-
PEX12 regulates PEX5 recycling by activating the E3 ubiquitin-protein ligase activity of PEX10; when recycling is compromised, it stimulates PEX10-mediated polyubiquitination of PEX5 for degradation (PubMed:24662292; ubiquitination stimulation by similarity). Related references: PMID:24662292
-
Peroxin-12 is an integral membrane protein localized to the peroxisome membrane.
-
Mutations in PEX12 are associated with peroxisome biogenesis disorders, including Zellweger syndrome spectrum.
-
The protein contains zinc-finger domains and transmembrane helices, indicating roles in metal binding and membrane integration.
-
The predicted molecular weight of PEX12 is approximately 40.8 kDa (UniProt), consistent with the product-reported 41 kDa.
Experimental Guidance and Technical Tips
- The immunogen used corresponds to the C-terminal region (amino acids 290-359) of human PEX12; this epitope selection may favor detection of the full-length protein.
- For Western blot, validate the antibody in human cell lines that express peroxisomal proteins, such as HepG2 or HeLa, and include appropriate positive and negative controls.
- For ELISA, consider using the recombinant immunogen as a reference antigen to establish standard curves and optimize coating conditions.
- As a polyclonal antibody, lot-to-lot variability is possible; it is advisable to verify each new lot with known standards.
- Cross-reactivity has been confirmed for human; for applications involving other species, experimental validation is recommended.
- Optimize working concentrations experimentally; typical starting dilutions for WB are often in the range of 1:500–1:2000, but this must be empirically determined.
