Biological Background: PERP Function and Localization

  • PERP (p53 apoptosis effector related to PMP-22), also known as keratinocyte-associated protein 1 (KCP-1), P53-induced protein PIGPC1, and transmembrane protein THW, is a component of intercellular desmosome junctions.
  • Plays a critical role in stratified epithelial integrity and cell-cell adhesion by promoting desmosome assembly, thereby contributing to the barrier function of the skin and mammary tissue homeostasis.
  • Required for tooth enamel development via facilitating desmosome-mediated ameloblast adhesion and may regulate downstream genes such as AMBN, ENAM, MMP20, and KLK4.
  • Serves as an effector in the TP53-dependent apoptotic pathway and positively regulates apoptosis in T-helper 17 (Th17) cell populations through caspase-dependent signaling.
  • Promotes neutrophil transepithelial migration in response to chemoattractants like HXA3, fMLP, and CXCL8, and is required for neutrophil migration during S. typhimurium infection. Related references: PMID:25486861
  • Localizes to the cell membrane, desmosome cell junctions, and cytoplasm.
