Biological Background: Peroxiredoxin-6 Function and Localization
- Peroxiredoxin-6 (PRDX6), also known as 1-Cys peroxiredoxin, AOP2, NSGPx, or aiPLA2, is a member of the peroxiredoxin family of antioxidant enzymes.
- It functions as a thiol-specific peroxidase, reducing hydrogen peroxide and organic hydroperoxides to water and alcohols, thereby protecting cells from oxidative damage. Related references: PMID:10893423, PMID:9497358
- PRDX6 can also reduce phospholipid hydroperoxides, playing a direct role in repairing oxidatively damaged cell membranes. Related references: PMID:10893423
- In addition to peroxidase activity, it possesses phospholipase A2 (PLA2) activity, hydrolyzing the sn-2 ester bond of phospholipids, which is regulated by pH and lipid binding. Related references: PMID:10893423, PMID:26830860

- The immunogen corresponds to amino acids 125-224 of human PRDX6, targeting the C-terminal region. This design is expected to recognize full-length PRDX6 in denaturing conditions suitable for Western blot.
- For Western blot, validate detection of a ~25 kDa band in whole cell lysates from human, mouse, or rat tissues, taking advantage of the reported cross-reactivity.
- For ELISA, consider using this monoclonal antibody as a capture or detection reagent after optimizing with recombinant PRDX6 protein or native samples.
