Biological Background: Peroxiredoxin 3 (PRDX3) Function and Localization
- Peroxiredoxin 3 (PRDX3) is a mitochondrial thioredoxin-dependent peroxide reductase, also known as Antioxidant protein 1 (AOP-1), MER5, and Prx-III.
- It functions as a thiol-specific peroxidase that detoxifies hydrogen peroxide and organic hydroperoxides, playing a critical role in cellular defense against oxidative stress. Related references: PMID:17707404 PMID:29438714 PMID:33889951
- PRDX3 acts synergistically with MAP3K13 to regulate NF-kappa-B activation in the cytosol. Related references: PMID:12492477
- It is predominantly localized to the mitochondrion but also detected in the cytoplasm and early endosomes.
- The protein undergoes multiple post-translational modifications, including acetylation, phosphorylation, and palmitoylation, and contains a redox-active center with a disulfide bond.
- Mutations in PRDX3 are associated with corneal dystrophy and neurodegenerative disorders.
Experimental Guidance and Technical Tips
- For Western blot (WB), a band of approximately 28 kDa is expected. Consider using reducing conditions to account for potential disulfide bonds, and validate detection in relevant cell or tissue lysates from human, mouse, or rat.
- For immunofluorescence/ICC, anticipate a primarily mitochondrial staining pattern. Optimize fixation and permeabilization protocols accordingly.
- The immunogen (aa 63–256) covers the central and C-terminal regions of PRDX3; it may recognize multiple splice variants if they include this region.
- As a polyclonal antibody, lot-to-lot consistency should be verified by titration in the intended assay.
- Cross-reactivity with mouse and rat enables comparative studies across these species. For ELISA, consider optimizing coating conditions and antibody concentration.
