Biological Background: Peroxiredoxin 2 (PRDX2) Function and Localization
- Peroxiredoxin-2 (PRDX2) is a member of the peroxiredoxin family of antioxidant enzymes, encoded by the PRDX2 gene (also known as NKEFB and TDPX1). It is also referred to as natural killer cell-enhancing factor B (NKEF-B), thiol-specific antioxidant protein (TSA), and thioredoxin peroxidase 1.
- PRDX2 functions as a thiol-specific peroxidase, catalyzing the reduction of hydrogen peroxide and organic hydroperoxides to water and alcohols, respectively, thereby protecting cells against oxidative stress.
- By detoxifying peroxides, PRDX2 also acts as a sensor of hydrogen peroxide-mediated signaling events and may participate in signaling cascades involving growth factors and tumor necrosis factor-alpha by regulating intracellular H2O2 concentrations.
- The protein primarily localizes to the cytoplasm, where it carries out its redox functions.
- PRDX2 undergoes various post-translational modifications, including acetylation, phosphorylation, and disulfide bond formation at its redox-active center, which may modulate its activity and interactions.
- The PRDX2 gene is subject to alternative splicing, potentially generating multiple isoforms with distinct functions or regulatory roles.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 1-100 of human PRDX2, targeting the N-terminal region. Ensure the antigen sequence is conserved in your species of interest; this antibody cross-reacts with mouse and rat PRDX2.
- For Western blot, a band at approximately 22 kDa is expected. Optimize antibody dilution using a known positive control sample and confirm band identity with a peptide competition assay if needed.
- For ELISA applications, consider using the recombinant protein or the immunizing peptide as a coating antigen to establish assay conditions and determine appropriate antibody concentrations.
- Because PRDX2 is an abundant cytoplasmic protein involved in redox regulation, sample preparation should minimize oxidation artifacts; include reducing agents in lysis buffer and consider running non-reducing vs reducing gels to detect redox-dependent modifications.
