Biological Background: Peroxiredoxin 1 Function and Localization
- Peroxiredoxin-1 (PRDX1), also designated Natural killer cell-enhancing factor A (NKEF-A), Proliferation-associated gene protein (PAG), and Thioredoxin peroxidase 2, is a thiol-specific peroxidase belonging to the peroxiredoxin family.
- Catalyzes the reduction of hydrogen peroxide and organic hydroperoxides to water and alcohols, thereby protecting cells against oxidative stress.
- Functions as a sensor of hydrogen peroxide-mediated signaling events and participates in growth factor and tumor necrosis factor-alpha signaling cascades by modulating intracellular H₂O₂ levels. Related references: PMID:9497357
- Predominantly localizes to the cytoplasm and is also detected in melanosomes.
- Undergoes multiple post-translational modifications including acetylation, phosphorylation, disulfide bond formation, isopeptide bond linkage, and ubiquitination.
- Contains a conserved redox-active cysteine residue that forms a disulfide bond during the catalytic cycle, essential for its peroxidase activity.
- Plays a role in postmitotic motor neuron differentiation by reducing an intramolecular disulfide bond in GDPD5 (By similarity).
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 99–199 of human Peroxiredoxin 1 (NP_001189360.1). This region is expected to be specific; however, perform appropriate controls to confirm specificity in your sample system.
- This antibody is validated for indirect immunofluorescence (IF/ICC) and ELISA. For IF/ICC, optimize fixation and permeabilization steps. For ELISA, consider using the recombinant protein or peptide as a positive control.
- While the predicted molecular weight is 22 kDa, performance in western blotting has not been listed. If you intend to use it for WB, we recommend pilot validation under your experimental conditions.
- The antibody is reported to cross-react with human, mouse, and rat Peroxiredoxin 1. Confirm reactivity for other species as needed.
