Biological Background: Peroxiredoxin 1/PAG Function and Localization

  • Also known as Natural killer cell-enhancing factor A (NKEF-A), Proliferation-associated gene protein (PAG), Thioredoxin peroxidase 2, and Thioredoxin-dependent peroxide reductase 2. It belongs to the peroxiredoxin family and is encoded by the PRDX1 gene in humans.
  • Functions as a thiol-specific peroxidase that reduces hydrogen peroxide and organic hydroperoxides to water and alcohols, playing a critical role in cellular antioxidant defense.
  • Acts as a sensor of hydrogen peroxide-mediated signaling events and may participate in growth factor and tumor necrosis factor-alpha signaling cascades by regulating intracellular H2O2 concentrations.

  • Localizes primarily to the cytoplasm and also to melanosomes, as indicated by subcellular localization data.
  • Post-translationally modified by acetylation, phosphorylation, and ubiquitin-like conjugation, and features a redox-active center with disulfide bond formation, essential for its catalytic cycle.
