Biological Background: PERK (EIF2AK3) Function and Localization
- EIF2AK3/PERK is a metabolic-stress sensor kinase that, upon ER stress, autophosphorylates and phosphorylates eIF2-alpha (EIF2S1) at Ser51, attenuating global protein synthesis while inducing ATF4-mediated integrated stress response (ISR) gene expression.

- For Western blot, protein lysates should be prepared under reducing conditions; a band near 125 kDa is expected. Low basal PERK expression may require ER stress induction with agents like tunicamycin or thapsigargin.
- In immunofluorescence and immunohistochemistry, consider co-staining with an ER marker (e.g., Calnexin) to confirm localization.
- For ELISA, consider using recombinant PERK protein or cell lysates from stressed cells as the capture/detection system.
