Biological Background: Periostin Function and Localization
- Periostin is a secreted extracellular matrix protein involved in cell adhesion, inducing cell attachment and spreading. Related references: PMID:12235007
- Enhances incorporation of BMP1 into the fibronectin matrix, promoting proteolytic activation of lysyl oxidase LOX, contributing to connective tissue integrity.
- Localized to the Golgi apparatus and secreted into the extracellular space and matrix.
- Widely expressed with highest levels in aorta, stomach, lower gastrointestinal tract, placenta, uterus, thyroid, and breast; also expressed in kidney and lung.
- Up-regulated in epithelial ovarian tumors, lung carcinoma periphery, and overexpressed in breast cancers; absent in normal ovaries.
- Alternative splicing produces multiple isoforms with molecular weights ranging from 80 to 93 kDa.
- Contains gamma-carboxyglutamic acid domains and heparin-binding sites, and is a glycoprotein with disulfide bonds.
Experimental Guidance and Technical Tips
- For Western blotting, consider using reducing conditions and a broad-range molecular weight marker to account for the multiple isoforms (80–93 kDa) and potential glycosylation.
- Validate the antibody in relevant human tissue or cell line lysates (e.g., aorta, breast cancer lines) to confirm specificity.
- For ELISA, consider coating with the immunogen peptide or recombinant Periostin, and optimize the antibody concentration for best signal-to-noise ratio.
- Due to the antibody's human specificity, it is not recommended for rodent samples unless cross-reactivity is independently confirmed.
