Biological Background: PER2 Function and Localization
- PER2 is a transcriptional repressor and core component of the circadian clock, generating approximately 24-hour rhythms in gene expression that regulate metabolism, sleep, body temperature, and immune function.
- It functions in the transcription/translation feedback loop (TTFL) by forming a negative limb: together with CRY1/2, it inhibits CLOCK/NPAS2-BMAL1/BMAL2 heterodimer activity, repressing its own expression.
- PER2 also recruits chromatin-modifying complexes (SIN3-HDAC, H3K9 methyltransferases) to target gene promoters and termination sites, regulating transcription initiation and elongation.
- Subcellularly, PER2 localizes to the nucleus, cytoplasm, perinuclear region, and nucleolus, shuttling as part of clock regulation.
- Post-translational modifications including phosphorylation, acetylation, and ubiquitination tune PER2 stability and circadian period length.
- Widely expressed with highest levels in skeletal muscle and pancreas; isoform 2 detected in keratinocytes. Mutations in PER2 cause familial advanced sleep phase syndrome (FASPS).
