Biological Background: PER1 Function and Localization

• PER1 is a core component of the circadian clock, acting as a transcriptional repressor in the transcription/translation feedback loop (TTFL) that generates ~24-hour rhythms.
• Together with PER2, PER3 and cryptochromes CRY1/2, it forms the negative limb of the circadian feedback loop, inhibiting the CLOCK/BMAL1 heterodimer.
• The circadian clock regulates wide-ranging physiological processes including metabolism, sleep, immune, cardiovascular, and renal functions.
• PER1 shuttles between the nucleus and cytoplasm (SUBCELLULAR LOCATION: Nucleus, Cytoplasm).
• It is subject to post-translational modifications (keyword: Phosphoprotein, Ubl conjugation) that determine the period (tau) of circadian rhythms.

• In immunofluorescence/ICC, PER1 localizes to both the nucleus and cytoplasm; expect a mixed subcellular staining pattern.
• For ELISA applications, consider coating with the recombinant immunogen or full-length PER1 protein for capture detection.
