Biological Background: PER1 Function and Localization
- PER1 is a core component of the circadian clock, acting as a transcriptional repressor in the transcription/translation feedback loop (TTFL) that generates ~24-hour rhythms.
- Together with PER2, PER3 and cryptochromes CRY1/2, it forms the negative limb of the circadian feedback loop, inhibiting the CLOCK/BMAL1 heterodimer.
- The circadian clock regulates wide-ranging physiological processes including metabolism, sleep, immune, cardiovascular, and renal functions.
- PER1 shuttles between the nucleus and cytoplasm (SUBCELLULAR LOCATION: Nucleus, Cytoplasm).
- It is subject to post-translational modifications (keyword: Phosphoprotein, Ubl conjugation) that determine the period (tau) of circadian rhythms.
- PER1 is widely expressed, with highest levels in skeletal muscle; found also in heart, brain, liver, kidney, and other tissues.
- Beyond circadian maintenance, PER1 modulates neuroinflammation, renal sodium reabsorption, hair follicle cycling, and glucocorticoid receptor activity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 207–275 of human PER1, so the antibody likely binds an epitope within this region.
- For Western blotting, bands around 136 kDa (full-length) and 92 kDa (possibly an isoform or cleavage product) may be observed; validate with appropriate controls.
- In immunofluorescence/ICC, PER1 localizes to both the nucleus and cytoplasm; expect a mixed subcellular staining pattern.
- For ELISA applications, consider coating with the recombinant immunogen or full-length PER1 protein for capture detection.
- As PER1 expression is circadian, sample collection timing should be carefully controlled to avoid variability in detection.
