Biological Background: PEPCK/PCK2 Function and Localization
- Encoded by the PCK2 gene, also known as PEPCK2 or PEPCK-M, this mitochondrial protein functions as a phosphoenolpyruvate carboxykinase (PEPCK) and a serine/threonine-protein kinase.
- Catalyzes the conversion of oxaloacetate to phosphoenolpyruvate, the rate-limiting step in gluconeogenesis and glyceroneogenesis, using GTP as a phosphate donor (PubMed:28955899). Related references: PMID:28955899
- Its kinase activity phosphorylates and activates ACSL4, thereby promoting ferroptosis, a form of regulated cell death (PubMed:38720107). Related references: PMID:38720107
- Belongs to the PEPCK family of carboxykinases, which play critical roles in energy metabolism and glucose homeostasis.
- Localizes exclusively to the mitochondrion, where it integrates into metabolic pathways of the citric acid cycle.
- Expressed widely across multiple tissues, consistent with its housekeeping metabolic functions.
- Annotated with diverse keywords including Acetylation, Alternative splicing, Decarboxylase, Lyase, Manganese, Metal-binding, Nucleotide-binding, and Transit peptide, indicating complex regulation and enzymatic properties.
- Molecular weight is approximately 71 kDa, but may vary due to post-translational modifications or alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 550-640); the antibody may detect both full-length and truncated forms of PCK2, but validation in relevant sample systems is recommended.
- For Western blot, consider using human, mouse, or rat cell lysates or mitochondrial fractions as positive controls; a band near 71 kDa is expected.
- For immunofluorescence or ICC, mitochondrial staining patterns should be confirmed with organelle-specific markers.
- Due to KO validation, the antibody can be employed in knockout/rescue experiments to demonstrate target specificity.
- ELISA protocols may require immobilization of recombinant PCK2 or capture of the endogenous protein from lysates; perform titration under your assay conditions.
