- PEN2, also known as presenilin enhancer protein 2, is a 101-amino acid, ~12 kDa transmembrane protein essential for gamma-secretase activity.

- PEN2 modulates presenilin endoproteolysis and is required for the maturation and activity of the gamma-secretase complex.
- The gamma-secretase complex plays a role in Notch and Wnt signaling pathways by regulating the processing of key regulatory proteins and cytosolic CTNNB1 levels.
- Beyond gamma-secretase, PEN2 acts as a regulator of AMP-activated protein kinase (AMPK) activity in response to calorie restriction, binding metformin and promoting V-ATPase complex inhibition.
