- Peflin (PEF1) is a 30 kDa calcium-binding protein belonging to the penta-EF-hand (PEF) family, also known as PEF protein with a long N-terminal hydrophobic domain.
- It functions as a Ca²⁺-dependent adapter that bridges unrelated proteins or stabilizes weak protein-protein complexes.
- In complex with PDCD6, PEF1 mediates ER-Golgi transport by regulating COPII coat size: the heterodimer bridges the BCR(KLHL12) complex to SEC31, promoting SEC31 monoubiquitination and collagen export (PubMed:27716508). Related references: PMID:27716508
- Conflicting evidence suggests PEF1 may also act as a negative regulator of PDCD6; some studies show calcium-induced dissociation of PEF1 from PDCD6, possibly inhibiting PDCD6-SEC31 interaction (PubMed:11278427). Related references: PMID:11278427
