Biological Background: PEDF/SERPINF1 Function and Localization
- PEDF/SERPINF1 (Pigment epithelium-derived factor) is a secreted glycoprotein and member of the serpin superfamily, also known as EPC-1 or PIG35. Unlike typical serpins, it does not undergo the stressed-to-relaxed conformational change and thus lacks serine protease inhibitory activity.
- PEDF functions as a potent neurotrophic factor, inducing extensive neuronal differentiation in retinoblastoma cells, and as a strong angiogenesis inhibitor.
- The protein is primarily secreted and has been detected in melanosomes. It is predominantly expressed in retinal pigment epithelial cells and blood plasma.
- Post-translational modifications include glycosylation and phosphorylation, with pyroglutamic acid at the N-terminus.
- PEDF has been implicated in conditions such as osteogenesis imperfecta and certain dwarfisms, as indicated by genetic disease associations.
- The mature protein has a molecular weight of ~46 kDa (calculated from amino acid sequence ~46.3 kDa).
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 1-100 of human PEDF/SERPINF1 (P36955). For WB and ELISA applications, consider using standard denaturing and non-denaturing conditions, respectively, and validate antibody specificity using appropriate positive and negative controls (e.g., recombinant PEDF, cell lysates from human, mouse, or rat origin).
- Since the antibody cross-reacts with mouse and rat, it is suitable for studies involving these species. However, confirm target band at ~46 kDa in your specific sample system.
- For ELISA, optimize coating antigen and antibody dilution; consider using an IgG control to ensure specificity.
- Given PEDF is secreted, cell culture supernatants or serum/plasma may be used as sample sources; be aware of potential interfering substances.
