Biological Background: PDZK1 Function and Localization
- PDZK1 (also known as NHERF3, CAP70, or NaPi-Cap1) is a PDZ domain-containing scaffolding protein that belongs to the Na(+)/H(+) exchange regulatory factor family.
- It serves as a scaffold connecting plasma membrane proteins and regulatory components, regulating their surface expression in the apical domains of epithelial cells.
- PDZK1 coordinates diverse regulatory processes for ion transport and second messenger cascades; in complex with NHERF1, it clusters functionally dependent proteins to modulate trafficking and activity.
- It plays a role in multidrug resistance through interaction with ABCC2 and PDZK1IP1, and potentiates the chloride channel activity of CFTR.
- PDZK1 is required for normal cell-surface expression of SCARB1, thereby helping to maintain plasma cholesterol levels, and it ensures proper localization and function of the chloride-anion exchanger SLC26A6 in the kidney proximal tubule.
- The protein localizes to the cell membrane and is expressed specifically in epithelial cells of the kidney (brush border of proximal tubule), pancreas, liver, and small intestine, with lower levels in the adrenal cortex, testis, and stomach; it is overexpressed in several carcinomas.
- PDZK1 is a phosphoprotein and may exist in multiple isoforms due to alternative splicing.
