Biological Background: PDX1 Function and Localization
- PDX1 (Pancreas/duodenum homeobox protein 1), also known as IPF1, IUF1, IDX-1, STF-1, GSF, is a homeobox transcription factor essential for pancreatic development and beta-cell function.
- Core transcriptional activator of insulin, somatostatin, glucokinase, islet amyloid polypeptide, and glucose transporter type 2 genes; critically involved in glucose-dependent regulation of insulin gene transcription.
- In pancreatic acinar cells, forms a complex with PBX1b and MEIS2b to activate the ELA1 enhancer, cooperating with the PTF1 complex bound to the enhancer A element.
- Binds preferentially to the DNA motif 5'-[CT]TAAT[TG]-3'.
- During embryonic development, specifies the early pancreatic epithelium, promoting proliferation, branching, and differentiation; in adults, maintains the hormone-producing phenotype of beta-cells.
- Subcellular localization is primarily nuclear, but also detected in the cytoplasm (cytosol).
- Tissue expression is restricted to duodenum and pancreas (Langerhans islet beta cells and small subsets of endocrine non-beta-cells, with low levels in acinar cells).
- The theoretical molecular weight is approximately 30.8 kDa; on SDS-PAGE, the protein typically migrates around 31 kDa.
