Biological Background: PDLIM5 Function and Localization

• Encodes PDZ and LIM domain protein 5 (also known as Enigma homolog, ENH), a scaffold protein that contains PDZ and LIM domains.
• In the heart, scaffolds protein kinase C to the Z-disk region, regulating cardiomyocyte expansion; overexpression promotes cardiac hypertrophy.
• Isoforms lacking LIM domains may negatively modulate the scaffolding activity of isoform 1; isoform 1 favors dendritic spine thinning and elongation in neurons.
• Localizes to the postsynaptic density, presynapse, postsynapse, cytoplasm, and cytosol.
• Tissue-specific expression in heart and skeletal muscle; commonly increased in brains of patients with bipolar disorder, schizophrenia, and major depression.
• Post-translational modifications include acetylation, phosphorylation, and ubiquitination; also contains metal-binding LIM domains.
• Gene: PDLIM5 (ENH); alternative splicing produces multiple isoforms.

Experimental Guidance and Technical Tips

• The immunogen corresponds to amino acids 317-596 of human PDLIM5, covering part of the C-terminal region. This region includes the PDZ domain, which may be conserved; thus, cross-reactivity with mouse and rat is supported.
• For Western blot (WB), a band at approximately 64 kDa is expected; however, multiple isoforms or post-translational modifications may result in additional bands. Validate in relevant cell or tissue lysates.
• For ELISA, use the antibody as a capture or detection reagent after confirming specificity with recombinant protein or known standards.
• Consider using heart or skeletal muscle tissues as positive controls, given the high expression in these tissues.
