Biological Background: PDLIM4 Function and Localization

  • PDLIM4, also known as RIL (Reversion-induced LIM protein), is a PDZ and LIM domain-containing protein that functions as a suppressor of SRC kinase activity.

  • It plays a key role in reorganization of the actin cytoskeleton by binding to alpha-actinin-1 (ACTN1), increasing its affinity for F-actin, and promoting stress fiber formation.

  • In neurons, PDLIM4 regulates synaptic AMPA receptor trafficking by linking internalized GRIA1-containing receptors to the actin cytoskeleton, thereby enhancing excitatory postsynaptic currents.
  • Upon oxidative stress, PDLIM4 binds to NQO1, which stabilizes it and leads to heterodimerization with isoform 1, attenuating cell migration.
  • The protein localizes to multiple subcellular compartments including the cytoplasm, cytoskeleton, nucleus, perinuclear region, lamellipodia, dendritic spines, and endosomal membranes (early and recycling endosomes).
