Biological Background: PDLIM4 Function and Localization
- PDLIM4, also known as RIL (Reversion-induced LIM protein), is a PDZ and LIM domain-containing protein that functions as a suppressor of SRC kinase activity.
Related references: PMID:19307596 - It plays a key role in reorganization of the actin cytoskeleton by binding to alpha-actinin-1 (ACTN1), increasing its affinity for F-actin, and promoting stress fiber formation.
Related references: PMID:21636573 - In neurons, PDLIM4 regulates synaptic AMPA receptor trafficking by linking internalized GRIA1-containing receptors to the actin cytoskeleton, thereby enhancing excitatory postsynaptic currents.
- Upon oxidative stress, PDLIM4 binds to NQO1, which stabilizes it and leads to heterodimerization with isoform 1, attenuating cell migration.
- The protein localizes to multiple subcellular compartments including the cytoplasm, cytoskeleton, nucleus, perinuclear region, lamellipodia, dendritic spines, and endosomal membranes (early and recycling endosomes).
- It is predominantly expressed in the brain and contains LIM and PDZ domains, which are involved in protein-protein interactions and metal binding (zinc).
- Keywords: Alternative splicing, phosphoprotein, synapse, zinc-binding.
Experimental Guidance and Technical Tips
- When performing Western blot, consider using a suitable secondary antibody and appropriate loading controls. The predicted molecular weight is approximately 35 kDa, but actual band migration may vary depending on the gel system and post-translational modifications.
- For immunofluorescence or immunocytochemistry, mild fixation with 4% paraformaldehyde and permeabilization with 0.1% Triton X-100 may be considered. However, fixation and permeabilization conditions should be optimized for each cell type and target.
- As the antibody cross-reacts with mouse and rat, validate its specificity in the relevant model system before proceeding to critical experiments. Use appropriate positive and negative controls.
- The immunogen corresponds to a recombinant fragment covering amino acids 80-210; this region may be conserved across isoforms, potentially allowing detection of multiple PDLIM4 variants. Verify isoform specificity if needed.
