Biological Background: PDLIM1/CLP36 Function and Localization
- PDLIM1 (PDZ and LIM domain protein 1), also known as CLP-36, Elfin, or CLIM1, is an adapter protein that links other proteins to the cytoskeleton. Related references: PMID:10861853
- It is involved in the assembly, disassembly, and directioning of stress fibers in fibroblasts, thereby regulating cytoskeletal dynamics.
- PDLIM1 is required for the proper localization of ACTN1 (α-actinin-1) and PALLD (palladin) to stress fibers, and for cell migration and maintenance of cell polarity.
- The protein localizes to the cytoplasm, cytoskeleton, and specifically to the Z lines of sarcomeres in myofibrils.
- It contains an N-terminal PDZ domain and a C-terminal LIM domain; the LIM domain coordinates zinc ions and mediates protein-protein interactions.
- Post-translational modifications include acetylation and phosphorylation, which may regulate its activity.
- PDLIM1 is strongly expressed in heart and skeletal muscle, moderately in spleen, small intestine, colon, placenta, and lung, and at lower levels in several other tissues; it is absent from brain, testis, ovary, and peripheral blood leukocytes.
Experimental Guidance and Technical Tips
- The immunogen used to generate this antibody corresponds to the full-length human PDLIM1 protein (amino acids 1-329), which may favor detection of both native and denatured protein forms.
- For Western blotting, the predicted molecular weight is approximately 36kDa; consider using heart or skeletal muscle lysates as positive controls due to high PDLIM1 expression.
- In immunofluorescence/immunocytochemistry, expect staining at stress fibers and Z lines; optimize fixation and permeabilization conditions for the specific cell type.
- The antibody is validated for ELISA; it may be suitable for antigen capture or detection in immunoassays.
- Cross-reactivity with mouse and rat has been confirmed, enabling studies across these species.
