Biological Background: PDLIM1/CLP36 Function and Localization

  • PDLIM1 (PDZ and LIM domain protein 1), also known as CLP-36, Elfin, or CLIM1, is an adapter protein that links other proteins to the cytoskeleton. Related references: PMID:10861853
  • It is involved in the assembly, disassembly, and directioning of stress fibers in fibroblasts, thereby regulating cytoskeletal dynamics.
  • PDLIM1 is required for the proper localization of ACTN1 (α-actinin-1) and PALLD (palladin) to stress fibers, and for cell migration and maintenance of cell polarity.
  • The protein localizes to the cytoplasm, cytoskeleton, and specifically to the Z lines of sarcomeres in myofibrils.
  • It contains an N-terminal PDZ domain and a C-terminal LIM domain; the LIM domain coordinates zinc ions and mediates protein-protein interactions.
