Biological Background: PDK2 Function and Localization
- PDK2, also known as pyruvate dehydrogenase kinase isoform 2 or PDHK2, is a mitochondrial kinase encoded by the PDK2 gene in humans. It belongs to the PDK/BCKDK protein kinase family and is expressed in many tissues, with highest levels in heart and skeletal muscle.
- Its core function is to phosphorylate and inactivate the pyruvate dehydrogenase complex (PDC) subunits PDHA1 and PDHA2, thereby regulating the conversion of pyruvate to acetyl-CoA and linking glycolysis to the tricarboxylic acid cycle.
- By inhibiting PDC activity, PDK2 down-regulates aerobic respiration and shifts cellular metabolism towards fatty acid oxidation, playing a key role in metabolic flexibility and adaptation to nutrient availability.
- PDK2 mediates cellular responses to insulin and helps maintain normal blood glucose levels and pH balance, preventing ketone body accumulation during starvation.
- The protein localizes to the mitochondrial matrix, where it directly interacts with the pyruvate dehydrogenase complex.
- Post-translationally, PDK2 is a phosphoprotein; its kinase activity is regulated by various metabolic signals. Alternative splicing produces multiple isoforms.

Experimental Guidance and Technical Tips
- The immunogen corresponds to a region within human PDK2 (aa 128-407). For Western blot (WB), consider using lysates from tissues with high PDK2 expression, such as heart or skeletal muscle. A band of approximately 46 kDa is expected.
- For immunofluorescence/immunocytochemistry (IF/ICC), mitochondrial localization should be observed; include appropriate mitochondrial markers as colocalization controls.
- In ELISA applications, the antibody may be used as a capture or detection reagent; optimization of coating concentration and detection conditions is recommended.
- Validate the antibody's reactivity in your specific sample system and application, performing necessary positive and negative controls.
