Biological Background: PDK2 Function and Localization
- PDK2 (pyruvate dehydrogenase kinase isoform 2) is a mitochondrial kinase encoded by the PDK2 gene. Multiple alternatively spliced isoforms have been reported.
- It phosphorylates and inactivates the pyruvate dehydrogenase complex (subunits PDHA1 and PDHA2), thereby regulating glucose and fatty acid metabolism, controlling the tricarboxylic acid cycle and aerobic respiration.
- PDK2 mediates cellular responses to insulin and plays a key role in maintaining normal blood glucose levels and in metabolic adaptation to nutrient availability.
- By regulating pyruvate dehydrogenase activity, PDK2 helps maintain normal blood pH and prevents the accumulation of ketone bodies under starvation conditions.
- It is involved in the regulation of cell proliferation, resistance to apoptosis induced by oxidative stress, and p53/TP53-mediated apoptosis.
- Subcellular localization: predominantly in the mitochondrial matrix.
- Tissue expression: highest in heart and skeletal muscle, intermediate in brain, kidney, pancreas and liver, and low in placenta and lung.

- The immunogen corresponds to the C-terminal region (aa 308-407) of human PDK2, a segment that may be conserved across species, consistent with the observed cross-reactivity with mouse and rat.
- In Western blotting, expect a major band around 46 kDa and a possible additional band at 39 kDa, reflecting different isoforms or processing variants; validate with appropriate positive and negative controls.
- For immunohistochemistry (IHC-P), consider standard antigen retrieval procedures and include tissue-specific controls, especially heart and skeletal muscle where PDK2 is highly expressed.
- For ELISA applications, the antibody can be tested as a capture or detection reagent; optimal working concentrations should be determined experimentally in the relevant matrix.
