Biological Background: PDHB Function and Localization

  • Encodes the beta subunit of pyruvate dehydrogenase E1 component (also known as PDHBD, PHE1B, or E1beta), which together with PDHA1 forms the heterotetrameric E1 subcomplex of the mitochondrial pyruvate dehydrogenase (PDH) complex.
  • Catalyzes the thiamine pyrophosphate (TPP)-dependent decarboxylation of pyruvate and the reductive acetylation of lipoyl groups on the E2 component, linking cytoplasmic glycolysis to the mitochondrial tricarboxylic acid (TCA) cycle (PubMed:17474719, PubMed:19081061).
    Related references: PMID:17474719, PMID:19081061
  • Resides in the mitochondrial matrix as a soluble enzyme, where it participates in carbohydrate and glucose metabolism.
  • Belongs to the pyruvate dehydrogenase E1 beta subunit family; mutations in the PDHB gene are associated with pyruvate dehydrogenase deficiency (a disease variant) and metabolic disorders.
  • Subject to post-translational modifications including phosphorylation and acetylation, which regulate enzyme activity in response to metabolic states.
  • Alternative splicing produces multiple transcript variants (as indicated by the keyword "Alternative splicing"), potentially contributing to functional diversity.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 1-300 of human PDHB (full-length), covering the mature protein region. Evaluate this design for detection of isoforms or splice variants if alternative splicing is a consideration.
  • This antibody is recommended for Western blot (WB) and ELISA applications. For WB, a band at approximately 39 kDa is expected; consider loading appropriate mitochondrial extracts or enriched samples to maximize signal.
  • Cross-reactivity has been observed with human, mouse, and rat PDHB; when working with other species, verify sequence homology of the immunogen region to predict potential reactivity.
  • As with any polyclonal antibody, lot-to-lot variation may occur. It is advisable to validate the antibody in the relevant sample system and optimize dilution factors for each experimental setup.
