Biological Background: PDHA2 Function and Localization
- Pyruvate dehydrogenase E1 component subunit alpha, testis-specific form, mitochondrial (PDHE1-A type II) is encoded by the Pdha2 gene in mouse.
- Together with PDHB, it forms the heterotetrameric E1 subunit of the pyruvate dehydrogenase (PDH) complex, catalyzing the conversion of pyruvate to acetyl-CoA and CO2, linking glycolysis to the TCA cycle.
- The E1 subunit catalyzes both the TPP-dependent decarboxylation of pyruvate and the reductive acetylation of a lipoyl group on the E2 component.
- Localizes to the mitochondrial matrix.
- Specifically expressed in testis.
- Participates in carbohydrate metabolism, glucose metabolism, and the tricarboxylic acid cycle.
- Requires magnesium and thiamine pyrophosphate as cofactors; subject to phosphorylation.
- The mature protein has a molecular weight of approximately 43 kDa (calculated from sequence: 43413 Da).
