Biological Background: PDGFRB Function and Localization
- Encodes the beta-type platelet-derived growth factor receptor, a tyrosine-protein kinase that acts as cell-surface receptor for homodimeric PDGFB and PDGFD and for heterodimers formed by PDGFA and PDGFB.
- Essential for embryonic development, regulating cell proliferation, survival, differentiation, chemotaxis, and migration.
- Critical for blood vessel development by promoting proliferation, migration, and recruitment of pericytes and smooth muscle cells to endothelial cells.
- Plays a role in vascular smooth muscle cell migration and neointima formation at vascular injury sites.
- Required for normal kidney glomerulus development, including pericyte (mesangial cell) recruitment and branched capillary network formation.
- Ligand binding activates multiple signaling cascades, including PLCG1, PI3K/AKT1, and RAS/MAPK pathways, leading to cytoskeletal rearrangement and membrane ruffle formation.
- Down-regulated by protein phosphatases and rapid internalization of the activated receptor.
- Alternative splicing produces multiple isoforms; implicated in various cancers and other diseases via chromosomal rearrangements and mutations.
Experimental Guidance and Technical Tips
- The immunogen used to generate this antibody targets a region within amino acids 200–300 of human PDGFRB, located in the extracellular domain. This region is predicted to be conserved across species, consistent with the reported cross-reactivity with mouse and rat.
- For western blot analysis, expect a band around 124 kDa for the full-length glycosylated receptor. Post-translational modifications and proteolytic processing may produce additional bands; validation in the relevant cell or tissue lysate is recommended.
- For immunohistochemistry and immunofluorescence, consider antigen retrieval methods and appropriate controls to confirm specificity.
- Although not tested for other applications, this polyclonal antibody may be suitable for immunoprecipitation or other assays with proper optimization.
