Biological Background: PDGFRB Function and Localization
- PDGFRB (Platelet-derived growth factor receptor beta, CD140B) is a receptor tyrosine kinase that binds PDGFB and PDGFD homodimers, as well as PDGFA/PDGFB heterodimers, initiating intracellular signaling cascades critical for cell proliferation, survival, differentiation, and migration.
- It plays an essential role in embryonic development and blood vessel formation by promoting proliferation, migration, and recruitment of pericytes and vascular smooth muscle cells to endothelial cells.
- PDGFRB can heterodimerize with PDGFRA, modulating downstream responses depending on the ligand and dimer composition.
- Upon ligand binding, the receptor autophosphorylates and activates multiple signaling pathways, including PLCG1-calcium-PKC, PI3K-AKT, and RAS-MAPK cascades, by phosphorylating adaptor proteins such as SHC1, GRB2, PTPN11, and others.
- Subcellularly, PDGFRB is located at the cell membrane, and upon activation it is internalized into cytoplasmic vesicles and lysosomes where signaling is downregulated.
- The protein undergoes post-translational modifications including tyrosine phosphorylation, glycosylation, and ubiquitination.

- The antibody was raised against a peptide spanning the C-terminal intracellular region (aa 1000-1106) of human PDGFRB. It may detect the full-length, unprocessed receptor but not secreted or truncated forms lacking this domain.
- For Western blotting, a band of approximately 124 kDa is expected. Consider using reducing conditions and appropriate controls to confirm PDGFRB expression.
- In immunofluorescence/ICC, signal may be observed at the plasma membrane and in intracellular vesicles. Fixation and permeabilization methods should be optimized for the cell type.
- Cross-reactivity with mouse PDGFRB has been reported; however, validation in the specific experimental system is recommended.
