Biological Background: PDGFRa Function and Localization
- PDGFRa (Platelet-derived growth factor receptor alpha) is a tyrosine-protein kinase receptor for PDGFA, PDGFB and PDGFC, encoded by the PDGFRA gene in humans. It is also known as CD140A, PDGFR2, or alpha-type platelet-derived growth factor receptor.
- Upon ligand binding, PDGFRa dimerizes and activates multiple downstream signaling cascades including PI3K/AKT, RAS-MAPK, and PLCG1 pathways, regulating cell proliferation, survival, chemotaxis, and differentiation. - PDGFRa is a single-pass transmembrane protein located at the cell membrane, and also found in the Golgi apparatus and primary cilium. It contains immunoglobulin-like domains in its extracellular region and a split tyrosine kinase domain intracellularly.
- The receptor is widely expressed, particularly in fibroblasts, smooth muscle, heart, brain, and embryos, and is detected in platelets and colon tumors. Its expression is important in mesenchymal stem cell differentiation and wound healing.
- Key post-translational modifications include N-linked glycosylation, autophosphorylation on tyrosine residues upon activation, and ubiquitination, which regulate its signaling and downregulation. It is classified as a proto-oncogene, with mutations associated with cancers and developmental disorders (e.g., gastrointestinal stromal tumors).
- PDGFRa can form heterodimers with PDGFRB, modulating signal specificity and intensity depending on the ligand isoform. The receptor signaling is attenuated by phosphatases and receptor internalization.

- The immunogen covers the extracellular domain (aa 24-528), so this antibody is likely to recognize the extracellular region. Consider this when designing experiments, for example, in non-reducing conditions or for native protein detection.
- For Western blot (WB), the predicted protein weight is ~123 kDa (may appear as a band around 170-180 kDa due to glycosylation). Validate in the relevant sample system, as expression levels can vary.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), optimize antigen retrieval and permeabilization. Cross-reactivity with mouse and rat allows use in common model organisms.
