Biological Background: PDGFD Function and Localization

  • PDGFD, also known as Iris-expressed growth factor (IEGF) or spinal cord-derived growth factor B (SCDGF-B), is a secreted glycoprotein and member of the platelet-derived growth factor family.
  • It functions as a potent mitogen for mesenchymal cells and plays essential roles in embryonic development, cell proliferation, migration, survival, and chemotaxis.
  • PDGFD is involved in wound healing and angiogenesis, inducing macrophage recruitment, increased interstitial pressure, and blood vessel maturation.
  • The protein is expressed at high levels in the heart, pancreas, adrenal gland, and ovary, with lower expression in placenta, liver, kidney, prostate, testis, small intestine, spleen, and colon.
  • In the kidney, PDGFD is expressed by visceral epithelial cells of the glomeruli and in smooth muscle cells of arteries, arterioles, and vasa rectae; its expression is altered in chronic obstructive nephropathy.
  • PDGFD expression is increased in prostate carcinoma and is also detected in many ovarian, lung, renal, and brain cancer-derived cell lines, suggesting a role in tumorigenesis.
  • The protein is synthesized as a precursor with a signal peptide and undergoes proteolytic cleavage on pairs of basic residues to generate the active growth factor.
  • Keywords associated with PDGFD include alternative splicing, disulfide bond, glycoprotein, growth factor, mitogen, proto-oncogene, and secreted.

Experimental Guidance and Technical Tips

  • The antibody was raised against a peptide within the central region (aa 250-350) of human PDGFD, which may recognize both full-length and cleaved forms.
  • For Western blotting, a band corresponding to the full-length PDGFD is expected at approximately 43 kDa; consider using appropriate controls and validate in relevant cell or tissue lysates.
  • Due to cross-reactivity with mouse, this antibody can be used in mouse models; however, sequence alignment of the immunogen region between species is recommended.
  • For ELISA, consider coating with recombinant PDGFD or peptide antigen and titrate the antibody to determine optimal working concentration.
  • Because PDGFD is a secreted protein, analyzing conditioned media or serum samples may require concentration steps to achieve detectable levels.
