Biological Background: PDGFC Function and Localization
- PDGFC encodes platelet-derived growth factor C, also known as fallotein, spinal cord-derived growth factor (SCDGF), or VEGF-E.
- It is a growth factor essential for embryonic development, regulating cell proliferation, migration, survival, and chemotaxis, particularly of mesenchymal cells.
- PDGFC is critical for normal skeleton formation, craniofacial development, palate formation, and skin morphogenesis during embryogenesis.
- It plays key roles in wound healing (inflammation, proliferation, remodeling), angiogenesis, blood vessel development, and fibrotic processes.
- The protein contains a CUB domain with mitogenic activity in smooth muscle cells, suggesting functions beyond maintaining PDGF domain latency, and may have additional roles in the nucleus.
- Subcellular localization includes cytoplasm, cytosol, secreted forms, nucleus, cytoplasmic granules, and cell membrane.
- It is highly expressed in fallopian tube, vascular smooth muscle, kidney, breast, colon, retinal pigment epithelia, platelets, prostate, testis, and uterus; expression is elevated in uterine leiomyomata.
- Keywords: Alternative splicing, Cleavage on pair of basic residues, Disulfide bond, Glycoprotein, Growth factor, Mitogen, Ubl conjugation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 166-345) of human PDGFC, which includes the growth factor domain. The antibody may detect both full-length and cleaved forms.
- For Western blot, the predicted molecular weight is 39 kDa; however, post-translational modifications or cleavage may result in multiple bands. Validate in your target sample system.
- For ELISA, pair with a capture antibody if developing a sandwich assay, and optimize coating conditions using recombinant protein or native samples.
- Cross-reactivity with mouse and rat is reported; further validation is recommended for other species.
