Biological Background: PDGFB Function and Localization
- PDGFB, also known as PDGF-2, platelet-derived growth factor B chain, beta polypeptide, and proto-oncogene c-Sis, is a secreted growth factor encoded by the PDGFB gene (synonyms: PDGF2, SIS).
- It functions as a potent mitogen for mesenchymal cells, driving cell proliferation, migration, survival, and chemotaxis during embryonic development and tissue homeostasis. Related references: PMID:26599395
- PDGFB is essential for normal vascular development: it is required for the proliferation and recruitment of pericytes and vascular smooth muscle cells in the central nervous system, skin, lung, heart, and placenta, and for blood vessel and kidney glomeruli formation.
- It plays a critical role in wound healing, where it stimulates chemotaxis and proliferation of fibroblasts and smooth muscle cells.
- PDGFB signaling is modulated by forming heterodimers with PDGFA (platelet-derived growth factor subunit A), altering receptor binding and downstream effects.
- The protein is secreted and contains characteristic disulfide bonds and glycosylation, contributing to its stability and dimerization.
- Tissue expression is greatest in the heart, brain substantia nigra, placenta, and fetal kidney, with moderate levels in hippocampus, skeletal muscle, adult kidney, and lung.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 140–241 of human PDGFB, which should recognize both the precursor and mature forms of the protein.
- For Western blot (WB), a band of approximately 27 kDa is expected under reducing conditions. Under non-reducing conditions, dimeric forms may appear at higher molecular weights due to interchain disulfide bonds; consider comparing reducing vs. non-reducing conditions to distinguish monomeric and dimeric populations.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), antigen retrieval (e.g., heat-mediated citrate buffer) is recommended due to the secreted nature of PDGFB; permeabilization is required for intracellular detection.
- For ELISA, use a coating antigen corresponding to the immunogen region or recombinant PDGFB protein; perform a standard curve to determine optimal capture and detection antibody concentrations.
- Cross-reactivity is reported with human and rat; validate in mouse or other species if needed, as sequence homology may vary.
- Always include appropriate positive and negative controls (e.g., PDGFB-overexpressing cell lysate vs. knockout or tissue known to lack expression) to confirm specificity.
